Label-free quantitative measurement of starch granules in the chloroplast

Not only as a photoautotrophic organelle, but also the place where many other molecules are synthesized, chloroplast has attracted a great deal of interest as a topic on the field of plant biology. In studying chloroplasts, one major approach is to examine the morphology of chloroplast using various microscopic techniques. Recently, optical diffraction tomography (ODT) was introduced as a way of imaging the cellular structure. ODT results in 3D refractive index (RI) distribution of live cells by detecting diffraction of light at different angles. Since RI is characteristic value of materials, 3D RI map offers information about the materials composing the cell without any labeling. In this study, we were able to visualize chloroplast using ODT. Furthermore, using ODT, we were able to quantitatively compare chloroplast volume, mass, and volume of starch granule between chloroplasts from Col-0 and chloroplasts from glk1 glk2 mutant without any label. The volume of chloroplast from WT was twice as large as that from glk1 glk2. Also, the volume ratio of starch granule to the chloroplast was 16.26% in WT while it was 6.91% in glk1 glk2 mutant.
