A developmental lysine-ε-acetylome of Arabidopsis thaliana

Lysine-ε-acetylation (Kac) is a post-translational modification playing important roles in metabolic regulation and cell signaling. However, its prevalence and specific functions in plants remains rather unexplored. Using high-resolution tandem mass spectrometry (MS) we characterized the Kac in 5 representative tissues of Arabidopsis thaliana and identified 5,656 Kac proteins and 11,861 Kac sites with high confidence. Proteins targeted by Kac show lower-than-average tissue-specific expression but higher tissue-specific Kac regulation, indicating Kac is an important component of the developmental program. We found that Kac preferentially target proteins that are conserved in plants, animals and fungi, such as enzymes involved in the glycolysis and the pentose-phosphate pathway. In contrast, plant- or Arabidopsis-specific proteins are less likely to be targeted. Although the lysines targeted by Kac tend to be conserved during evolution, the Kac sites are not always conserved in different species. Besides mitochondria and nucleus, the energy-harvesting organelle chloroplast is also a main site of protein acetylation. Kac strongly interacts with other PTMs such as ubiquitination, SUMOylation and phosphorylation, but shares few common sites with ubiquitination or SUMOylation, which also target lysine. In summary this study revealed a comprehensive map of the lysine-ε-acetylome in the model plant Arabidopsis and suggests that Kac is dynamically regulated during plant development.
