High-resolution mapping of three-dimensional (3D) structures in intact tissues is indispensable in many biomedical studies. Optical imaging combined with fluorescence labelling techniques shows fast and simple choices for the 3D reconstruction of tissue structures, but the imaging depth is limited to the strong light scattering of tissues. Tissue optical clearing techniques provide a new way for large volume tissue imaging. No matter how, solvent-based clearing methods, or aqueous-based clearing methods, each has its advantages, and some limitations. In this presentation, we will demonstrate typical optical clearing imaging methods we developed, including advanced tissue optical clearing methods to preserve the fluorescence of various fluorescent probes, or allows rapid and efficient to clear kinds of tissues, and minimize tissue distortion for brain-wide profiling of diverse architectures. In addition, we developed a novel vascular labelling technique. Optical clear methods combined with fluorescence labelling techniques, promise us to map 3D structure of whole organs.
