Construction of nano drug delivery system based on marine polysaccharide and its anti-hepatic fibrosis effect

Liver fibrosis is a reversible over-repair reaction of liver tissue injury, which will progress to cirrhosis and even liver cancer if it is not treated promptly and effectively. Fucoidan, as one of the representative ingredients of seaweed, has been found to significantly inhibit the expression of TGF-β1/Smads, the key pathway of liver fibrosis, simultaneously inhibit the expression of tissue inhibitor of metalloproteinase -1(TIMP-1), promote the expression of metalloproteinase -9(MMP-9), and inhibit the process of liver fibrosis through antioxidation and anti-inflammation. In this paper, PH-sensitive multifunctional marine biological material natural polysaccharide nanoparticles were designed, which contained the effective components of traditional Chinese medicine. Baicalin (Bai) with fucoidan as hydrophilic end, imine bond as PH sensitive bond connected to hydrophobic end and folic acid (FA) modified hydrophilic end targeted liver cells.We prepared curcumin nanocrystals coated with amphiphilic carrier materials.Cur nanocrystals have the characteristics of high loading and good stability. Baicalin and curcumin can regulate TGF-β1/Smads signaling pathway to achieve anti-hepatic fibrosis effect, and play a synergistic role with fucoidan to achieve good anti-hepatic fibrosis effect.
